Safinamide + levodopa is a combination therapy used in the treatment of Parkinson's disease, specifically for patients experiencing "off" episodes—periods when symptoms such as difficulty moving, walking, and speaking return as medication effects wear off. Safinamide is a selective, reversible monoamine oxidase B (MAO-B) inhibitor that increases and prolongs dopamine activity in the brain by inhibiting its breakdown. In addition to MAO-B inhibition, safinamide also blocks voltage-dependent sodium and calcium channels and inhibits glutamate release, which may provide additional benefits in controlling motor symptoms and reducing dyskinesia. Levodopa is a dopamine precursor that replenishes depleted dopamine levels in the brain. This combination aims to optimize dopaminergic stimulation for improved symptom control in Parkinson's disease[1][2][4][5][6][8].
