Overview

Safrazine is a non-selective, irreversible monoamine oxidase inhibitor (MAOI) of the hydrazine class. It was introduced in the 1960s as an oral antidepressant but has since been discontinued. Safrazine inhibits both types of monoamine oxidase enzymes (MAO-A and MAO-B), leading to increased levels of neurotransmitters such as serotonin and phenylethylamine in the brain. Its action is characterized by long-lasting inhibition due to its irreversible binding, with effects on monoamine content persisting for at least 24 hours after administration. The drug was withdrawn from clinical use due to safety concerns and the development of newer antidepressants with improved efficacy and safety profiles[1][2][3][4][5][6][7].
