SAIL66 is a next-generation tri-specific monoclonal antibody designed as a T-cell engager (TCE) for the treatment of solid tumors expressing Claudin 6 (CLDN6). It binds to CLDN6 on tumor cells with one Fab arm and to both CD3 and CD137 on T cells with the other Fab arm. This dual engagement activates T cells through CD3 signaling and provides co-stimulation via CD137, resulting in enhanced antitumor activity compared to conventional CLDN6-targeting TCEs. Preclinical studies have shown that SAIL66 increases intratumoral T-cell infiltration, reduces exhausted T cell populations, and demonstrates high specificity for CLDN6 without binding to related claudin family members. Clinical trials are ongoing for ovarian cancer and other solid tumors expressing CLDN6.
