SAIT301 is a humanized IgG2 monoclonal antibody that specifically targets the alpha chain of the extracellular Sema domain of the c-MET (mesenchymal epithelial transition factor) receptor. Developed by the Samsung Advanced Institute of Technology, it functions by blocking the binding of the ligand hepatocyte growth factor (HGF), thereby inhibiting HGF-mediated oncogenic signaling. A key differentiator of SAIT301 is its ability to induce efficient c-MET internalization and subsequent lysosomal degradation through a Cbl-independent, LRIG1-mediated pathway. This unique mechanism enables it to act as a fully antagonistic antibody without the intrinsic agonistic activity often seen in other bivalent anti-MET antibodies. Clinical investigation has primarily focused on advanced c-MET-positive solid tumors, including metastatic colorectal cancer.
