Overview

Salatinib (QL1209) is an orally bioavailable, small-molecule irreversible dual tyrosine kinase inhibitor (TKI) developed by Qilu Pharmaceutical. It is designed to target both the Human Epidermal Growth Factor Receptor 2 (HER2) and the Epidermal Growth Factor Receptor (EGFR). By covalently binding to the intracellular kinase domains of these receptors, salatinib inhibits downstream signaling pathways, such as the MAPK and PI3K/Akt pathways, which are critical for the survival and proliferation of tumor cells that overexpress HER2 or EGFR. The drug is primarily being developed for the treatment of HER2-positive metastatic breast cancer, where it is often evaluated in combination with chemotherapy agents like capecitabine. Salatinib is currently in late-stage clinical development, including Phase 3 trials, aiming to provide a potent therapeutic option for patients who have progressed on prior HER2-targeted therapies.
