saline + succinylcholine is a pharmacological combination used as a control intervention in clinical research, specifically in a Phase 3 trial (NCT04709315) conducted by South Valley University to evaluate intubating conditions during rapid sequence induction (RSI). Succinylcholine, the active component, is a depolarizing neuromuscular blocking agent that acts as an agonist at nicotinic acetylcholine receptors (nAChR) at the neuromuscular junction. By mimicking acetylcholine but resisting degradation by acetylcholinesterase, it produces persistent depolarization of the motor endplate, leading to transient muscle fasciculations followed by skeletal muscle paralysis. In this specific trial protocol, a 0.9% saline infusion is administered as a 10-minute pretreatment to serve as a placebo control before the administration of succinylcholine (1 mg/kg) to facilitate tracheal intubation in patients undergoing elective surgery requiring rapid sequence induction.
