Overview

The vaccine consists of live-attenuated S. Typhi strains engineered to express the pneumococcal surface protein A (PspA). This approach utilizes the natural invasive properties of Salmonella to deliver the pneumococcal antigen to the gut-associated lymphoid tissue (GALT), thereby stimulating both mucosal and systemic immune responses. The specific strains (χ9088, χ9558, and χ8914) incorporate various genetic modifications to balance attenuation with immunogenicity, aiming to provide a cost-effective, orally administered alternative to conjugate vaccines with broader coverage across pneumococcal serotypes.
