Overview

Salmonella Typhimurium SL3261 is a live, attenuated bacterial strain derived from Salmonella enterica serovar Typhimurium SL1344 by deletion of the aroA gene, rendering it auxotrophic for aromatic compounds and significantly attenuated in virulence[3]. It is widely used as a vaccine vector and experimental immunotherapy platform. Mechanistically, it functions by entering host tissues, stimulating both mucosal and systemic immune responses, and serving as a live delivery vehicle for heterologous antigens or therapeutic molecules (e.g., recombinant proteins, shRNA-expressing plasmids)[2][4][5][6]. SL3261 elicits robust humoral and cellular immunity, and is being researched as an oral vaccine for infectious diseases (ex. Cryptosporidium parvum, Haemophilus ducreyi) and as an anticancer immunotherapy/tumor-targeting agent for localized delivery of therapeutic payloads[2][4][6]. SL3261 is not an approved pharmaceutical but is frequently used in preclinical models for proof-of-concept studies of novel vaccines and immunotherapies[2][5][6].
