Drug intelligence / Profile preview
salsalate + docosahexaenoic acid (DHA) is a combination of the nonsteroidal anti-inflammatory drug (NSAID) salsalate and the omega-3 fatty acid docosahexaenoic acid. This specific combination was utilized by Catabasis Pharmaceuticals as an active comparator arm in a Phase 1 crossover biomarker study (Study 103; NCT01670773) for the treatment of Duchenne muscular dystrophy (DMD). The study was designed to compare the NF-κB inhibitory effects of this unconjugated combination against edasalonexent (CAT-1004), which is a covalent conjugate of salicylic acid and DHA. Results from the trial demonstrated that while edasalonexent successfully inhibited NF-κB activity following a single dose, the equimolar combination of salsalate and DHA did not achieve significant inhibition, supporting the therapeutic rationale for the conjugated molecule over the simple combination of its components.
