SAM-315 (also known as WAY‑255315) is a small molecule drug that acts as a potent antagonist of the serotonin 6 (5‑HT6) receptor. It was initially developed by Wyeth and later Pfizer for the treatment of Alzheimer's disease and other nervous system disorders. The compound demonstrated high affinity for the 5‑HT6 receptor, with reported Ki and IC50 values of approximately 1.1 nM and 4.6 nM, respectively. Despite early clinical development—including multiple phase I trials to assess safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy subjects—development was discontinued after phase I due to unspecified reasons[1][2][3]. No approved indications or commercial formulations exist.
