SAM001 is an investigational small molecule drug developed by Samsara Therapeutics for the treatment of neurodegenerative diseases, specifically Parkinson’s disease and amyotrophic lateral sclerosis (ALS). It functions as an oral, once-daily autophagy enhancer that targets Transient Receptor Potential Mucolipin 1 (TRPML1), a protein involved in regulating autophagy. By activating TRPML1, SAM001 boosts the cell’s self-cleaning process to remove toxic protein build-up associated with these diseases. Preclinical studies have shown that SAM001 can cross the blood-brain barrier, improve nerve and muscle function, extend survival in ALS mouse models, reduce neuronal damage markers, and reverse disease symptoms without significant safety concerns. The drug was discovered using Samsara Therapeutics’ Lysoseeker platform[1][3][4][5].
