Samatatatug zovodotin (XB002) is a next-generation antibody-drug conjugate (ADC) targeting Tissue Factor (TF), also known as coagulation factor III. It is composed of a human monoclonal antibody (samatatatug) conjugated to a potent cytotoxic payload, zovodotin (an auristatin derivative), via a protease-cleavable linker. The drug is designed to bind to TF on the surface of tumor cells, leading to internalization and the subsequent release of the payload, which inhibits microtubule polymerization and induces apoptosis. Notably, samatatatug zovodotin is engineered to bind TF without interfering with the coagulation cascade, a design intended to improve the safety profile regarding bleeding compared to earlier TF-targeting ADCs. It is currently being evaluated in Phase 1 clinical trials for the treatment of various advanced solid tumors, including cervical, ovarian, and lung cancers.
