SAMHD1 siRNA is a small interfering RNA designed to silence the expression of the SAMHD1 (sterile alpha motif and HD domain-containing protein 1) gene. SAMHD1 is a deoxynucleoside triphosphate triphosphohydrolase (dNTPase) that regulates cellular dNTP pools and has been implicated in viral defense and cell cycle regulation. In the context of oncology, specifically clear cell renal cell carcinoma (ccRCC), SAMHD1 has been identified as an oncogene that promotes cell migration and metastasis. Mechanistically, SAMHD1 contributes to the formation of an endosomal complex by binding to cortactin, which stimulates endosomal focal adhesion kinase (FAK) signaling and activates Rac1, leading to lamellipodia formation. SAMHD1 siRNA is primarily used as a research tool to inhibit this pathway, thereby reducing the motility and invasive potential of cancer cells in preclinical models.
