Samuraciclib + chemotherapy is an investigational combination regimen developed by Carrick Therapeutics for the treatment of advanced solid tumors. The regimen consists of samuraciclib (CT7001), a first-in-class, oral small molecule inhibitor of Cyclin-Dependent Kinase 7 (CDK7), administered in conjunction with cytotoxic chemotherapy agents. CDK7 is a critical regulator of the cell cycle (via activation of CDKs 1, 2, 4, and 6) and gene transcription (via phosphorylation of RNA polymerase II). By inhibiting CDK7, samuraciclib suppresses the transcription of oncogenic drivers and disrupts uncontrolled cell cycle progression, which may sensitize tumor cells to the DNA-damaging effects of chemotherapy. This combination has received FDA Fast Track designation for locally advanced or metastatic triple-negative breast cancer (TNBC) and is being evaluated in Phase 1 clinical trials for pancreatic cancer.
