SAN2465 is a small molecule negative allosteric modulator (NAM) of the GABA-A receptor alpha-5 subunit, currently being developed through a collaboration between Saniona and Boehringer Ingelheim. It is primarily targeted for the treatment of major depressive disorder (MDD) and severe depression. By selectively modulating the alpha-5 subunit of the GABA-A receptor—which is highly expressed in brain regions like the hippocampus and prefrontal cortex—SAN2465 aims to reduce excessive inhibitory signaling, potentially offering rapid-acting antidepressant effects. The program has progressed into early clinical development, with pipeline reports indicating Phase 1 and Phase 2a stages of evaluation.
