Overview

SAN523 is a small molecule positive allosteric modulator (PAM) of cystathionine gamma-lyase (CSE), an enzyme responsible for the endogenous production of hydrogen sulfide (H2S). By enhancing CSE activity, SAN523 increases H2S levels, which in turn inhibits the activation of the NLRP3 inflammasome. This inhibition leads to reduced secretion of pro-inflammatory cytokines, specifically IL-1β. In preclinical studies, SAN523 has demonstrated the ability to reduce inflammatory cell recruitment and cytokine levels in mouse models of MSU-induced peritonitis, suggesting potential therapeutic application in acute gout and other IL-1β-mediated inflammatory conditions.
