Overview

SANA-236 is an investigational, hypoimmune (HIP) modified, allogeneic chimeric antigen receptor (CAR) T-cell therapy targeting GPRC5D, developed by Sana Biotechnology for the treatment of multiple myeloma. The therapy utilizes Sana's HIP platform, which involves the genetic disruption of the T-cell receptor (TCR), beta-2 microglobulin (B2M), and class II major histocompatibility complex transactivator (CIITA) to prevent recognition by the host's adaptive immune system. Additionally, it overexpresses CD47 to inhibit phagocytosis by innate immune cells, such as macrophages. By targeting GPRC5D, a protein highly expressed on malignant plasma cells but with limited expression in healthy tissues (primarily hair follicles and bone marrow), SANA-236 aims to provide a potent and persistent off-the-shelf treatment option for patients with relapsed or refractory multiple myeloma.
