Overview
The Sanofi protein-based pneumococcal vaccine (PPrV) is an investigational vaccine candidate developed by Sanofi Pasteur for the prevention of infections caused by *Streptococcus pneumoniae*. Unlike traditional pneumococcal conjugate vaccines (PCVs) that target the polysaccharide capsule of specific serotypes, the PPrV approach utilizes highly conserved surface proteins to provide broader, serotype-independent protection. The primary antigens in these formulations typically include the Pneumococcal histidine triad protein D (PhtD) and a detoxified version of pneumolysin (dPly). These proteins are essential for the virulence and colonization of the bacteria across most known serotypes. Sanofi has evaluated several versions of this vaccine, such as SP0125 and SP0148, in Phase 1 and Phase 2 clinical trials to assess their safety and ability to induce a robust immune response in healthy adults and pediatric populations.
