Overview

Sanquin Notch-engineered T-cell therapy is an experimental cell therapy platform developed by Sanquin Innovatie for the treatment of solid tumors. This approach utilizes synthetic Notch (synNotch) receptors, which are chimeric proteins designed to provide T-cells with sophisticated environmental sensing capabilities. In this system, the binding of a primary tumor-associated antigen to the synNotch receptor triggers the proteolytic cleavage and release of an internal transcription factor. This factor then migrates to the nucleus to drive the expression of a secondary therapeutic gene, such as a Chimeric Antigen Receptor (CAR) or a cytokine. By requiring two distinct antigens for full activation (an AND-gate logic), this technology aims to significantly enhance the tumor-specificity of T-cell therapies and mitigate the risks of off-tumor, on-target toxicity commonly associated with traditional CAR-T approaches in solid malignancies.
