Overview

**Sapanisertib + paclitaxel** is an investigational combination therapy comprising **sapanisertib**, a potent and selective dual inhibitor of mammalian target of rapamycin complexes 1 and 2 (mTORC1/2), and **paclitaxel**, a cytotoxic chemotherapeutic agent that stabilizes microtubules and prevents their depolymerization[5][6][7]. Sapanisertib blocks mTORC1 and mTORC2 signaling, crucial pathways driving cell growth, proliferation, and survival—especially in cancers with PI3K/mTOR/AKT pathway aberrations[5][6][7]. Paclitaxel, by interfering with microtubule function, induces cell cycle arrest and apoptosis. Preclinical and clinical studies suggest that combining sapanisertib with paclitaxel can sensitize tumor cells to taxane chemotherapy and overcome resistance mechanisms driven by hyperactive PI3K/mTOR signaling, thereby improving antitumor efficacy[5][6][7]. Development efforts are focused primarily on advanced solid tumors, particularly endometrial, ovarian, breast, and prostate cancers[5][6][7].
