Overview
SAR-405838 (also known as MI-77301) is a highly potent and selective small molecule inhibitor of the MDM2-p53 interaction. It binds to MDM2 with high affinity, preventing the binding of MDM2 to the tumor suppressor protein p53. This inhibition blocks proteasome-mediated degradation of p53, leading to restoration of p53 activity, which can induce cell cycle arrest or apoptosis in cancer cells with wild-type p53. SAR-405838 has demonstrated antitumor activity in preclinical models and was investigated in phase I clinical trials for advanced solid tumors and malignant neoplasms. The drug was initially developed by Sanofi[1][2][3][4][5][6].
