Overview

SAR228810 is a humanized monoclonal antibody developed for the treatment of Alzheimer's disease. It is designed to specifically bind to soluble protofibrillar and fibrillar forms of amyloid-β (Aβ) peptide, with minimal activity against Aβ monomers and small oligomeric aggregates. The antibody features limited Fc effector functions due to two silencing mutations on a human IgG4 backbone, aiming to reduce the risk of amyloid-related imaging abnormalities (ARIA) and improve safety compared to earlier anti-Aβ antibodies. By targeting pathology-associated Aβ species rather than monomers, SAR228810 seeks to prevent plaque formation and synaptic dysfunction more effectively. Clinical development has included phase 1 studies in patients with mild-to-moderate Alzheimer's disease[1][2][3][4][5][8].
