Overview
**SAR245408 + seribantumab** is an experimental combination therapy comprising SAR245408 (also known as pilaralisib), a small molecule inhibitor of phosphoinositide 3-kinase (PI3K), and seribantumab (MM-121), a fully human monoclonal antibody targeting the human epidermal growth factor receptor 3 (HER3/ErbB3). SAR245408 inhibits PI3K, leading to suppression of downstream signaling pathways involved in cell proliferation and survival. Seribantumab binds HER3, blocking heregulin-mediated activation and downregulating receptor signaling. The combination aims to target cancers with activation of the PI3K pathway or HER3 signaling, especially solid tumors with *PIK3CA* mutations or NRG1-HER3 dependence.
