Overview

SAR405 is a potent and selective small molecule inhibitor of the class III phosphoinositide 3-kinase (PIK3C3, also known as Vps34). By competitively binding to the ATP pocket of Vps34 (Kd: 1.5 nM; IC50: 1.2 nM), SAR405 inhibits its catalytic activity, which disrupts vesicle trafficking from late endosomes to lysosomes and prevents autophagy[1][3][5][7]. SAR405 also fully inhibits autophagy induced by starvation or MTOR (mechanistic target of rapamycin) inhibition and has shown synergy with MTOR inhibition in reducing cell proliferation in tumor cell models[1][5][7]. Its high selectivity profile makes it a unique research tool for dissecting autophagy mechanisms[5][7].
