SAR428926 is an antibody-drug conjugate (ADC) designed for the treatment of advanced solid tumors. It consists of a humanized monoclonal antibody targeting lysosome-associated membrane protein 1 (LAMP1), which is involved in cell-cell adhesion and migration. The antibody is conjugated via a disulfide-containing cleavable linker (N-succinimidyl-4-(2-pyridyldithio)butyrate, SPDB) to the cytotoxic maytansinoid DM4 payload. Upon binding to LAMP1-expressing tumor cells, the ADC is internalized and releases DM4 intracellularly, where it acts as a microtubule inhibitor to induce cell death. Preclinical studies have shown efficacy in various patient-derived xenograft models of colon, breast, lung, prostate, gastric and ovarian cancers expressing LAMP1[1][2][5]. Clinical development has included phase 1 trials in patients with advanced solid tumors[4].
