Overview

SAR440234 is a bispecific monoclonal antibody developed by Sanofi for the treatment of hematologic malignancies, specifically relapsed or refractory acute myeloid leukemia (AML), B-cell acute lymphoblastic leukemia (B-ALL), and high-risk myelodysplastic syndromes (HR-MDS). It functions as a T-cell engager by simultaneously binding to CD3 on T cells and CD123 on tumor cells, thereby redirecting cytotoxic T lymphocytes to target and kill malignant cells expressing CD123. The drug’s mechanism involves antibody-dependent cell cytotoxicity and stimulation of T lymphocytes. Clinical development reached phase 1/2 trials but was terminated due to sponsor decision.
