SAR441000 is an investigational mRNA-based immunotherapeutic agent developed for intratumoral administration in patients with advanced solid tumors, including melanoma. It consists of a saline-formulated mixture of four mRNAs encoding interleukin-12 single chain (IL-12sc), interferon alpha-2b (IFNα2b), granulocyte-macrophage colony-stimulating factor (GM-CSF), and interleukin-15 sushi domain (IL-15sushi). These cytokines are designed to stimulate local and systemic antitumor immune responses by promoting immune cell infiltration, T-cell activation, and the formation of immune memory. Preclinical studies demonstrated that SAR441000 could induce tumor regression and enhance the efficacy of checkpoint inhibitors. Clinical development included phase 1 trials as monotherapy and in combination with cemiplimab for advanced solid tumors; however, development was discontinued prior to completion of phase 1[1][3][4][6].
