SAR441236 is a genetically engineered trispecific broadly neutralizing monoclonal antibody designed to target HIV. It combines three specificities into one molecule, enabling it to bind simultaneously to three distinct sites on the HIV envelope glycoprotein—specifically, the CD4 binding site (VRC01-LS), the V1/V2 glycan region (PGDM1400), and the gp41 membrane-proximal external region (10E8v4-variant). This design allows for broad and potent neutralization of diverse HIV strains, with demonstrated activity against 98% of viruses in standard panels and protection in non-human primate models. The antibody is further engineered with Fc-region modifications for extended half-life. Developed collaboratively by Sanofi, ModeX Therapeutics, and the National Institute of Allergy and Infectious Diseases (NIAID), SAR441236 is being evaluated primarily for treatment and prevention of HIV infection. Phase 1 clinical trials have shown it to be safe, well-tolerated, and effective at reducing viral load in people living with HIV[2][3][7][8].
