SAR442085 is a next-generation Fc-engineered monoclonal antibody that targets the cell surface glycoprotein CD38. It was developed as an antineoplastic agent for the treatment of multiple myeloma. The drug exhibits enhanced affinity for activating Fcγ receptors (FcγRIIa and FcγRIIIa), resulting in improved natural killer (NK) cell-dependent antibody-dependent cellular cytotoxicity (ADCC) compared to earlier anti-CD38 antibodies such as daratumumab and isatuximab. Preclinical studies demonstrated superior in vitro and in vivo antimyeloma efficacy, but clinical development was discontinued during phase I trials due to strategic portfolio decisions unrelated to safety[1][2][3][4].
