SAR442257 is a trispecific monoclonal antibody designed as a T-cell engager that targets CD38, CD3, and CD28. By simultaneously binding to CD3 and CD28 on T cells, it provides co-stimulatory signals that enhance T cell activation. The additional targeting of CD38 directs these activated T cells toward tumor cells expressing this antigen, such as those found in multiple myeloma and non-Hodgkin lymphoma. SAR442257 can also bind directly to CD28 expressed on tumor cells, further enhancing its antitumor activity even when other anti-CD38 antibodies are present. This mechanism allows for potent cytotoxic responses against cancerous cells by leveraging the patient’s own immune system. The drug is being developed primarily for relapsed/refractory multiple myeloma (RRMM) and relapsed/refractory non-Hodgkin lymphoma (RR-NHL), with ongoing Phase 1 clinical trials[1][2][4][5][7].
