Overview

SAR443579 (also known as IPH6101 or SAR'579) is an investigational trifunctional natural killer (NK) cell engager antibody designed to target hematological malignancies. It is engineered to simultaneously bind the tumor-associated antigen Cluster of differentiation 123 (CD123) on malignant cells and co-engage the activating receptors Natural cytotoxicity triggering receptor 1 (NKp46) and Fc gamma receptor III-A (CD16a) on NK cells. This tri-specific engagement facilitates the formation of a cytolytic synapse between NK cells and CD123-positive tumor cells, leading to selective activation of NK cells and subsequent tumor cell lysis. The drug is being developed by Sanofi in collaboration with Innate Pharma using Innate's proprietary ANKET multispecific antibody platform. Its primary indications under investigation are relapsed or refractory acute myeloid leukemia (R/R AML), B-cell acute lymphoblastic leukemia (B-ALL), high-risk myelodysplastic syndrome (HR-MDS), and blastic plasmacytoid dendritic cell neoplasm (BPDCN). Clinical trials have shown that SAR443579 has been well tolerated up to 3000 µg/kg weekly with observed clinical benefit in R/R AML patients[1][2][6][7].
