SAR443579 + azacitidine + venetoclax is an experimental three-drug combination regimen under investigation for the treatment of newly diagnosed, CD123-expressing acute myeloid leukemia (AML) in adults ineligible for intensive chemotherapy. **SAR443579** is a trifunctional natural killer cell engager (NKCE) designed to co-engage CD123 on tumor cells and the NK cell receptors NKp46 and CD16a, thereby forming a cytolytic synapse to activate NK cells and mediate targeted cell killing. **Azacitidine** is a hypomethylating agent (DNA methyltransferase inhibitor) that inhibits DNA methylation and induces cytotoxicity in abnormal hematopoietic cells. **Venetoclax** is a selective BCL-2 inhibitor that promotes apoptosis in cancer cells. The aim of combining these modalities is to increase anti-leukemic activity and overcome resistance mechanisms seen with venetoclax/azacitidine alone. This combination is currently in Phase 1/2 clinical trials sponsored by Sanofi, with SAR443579 being developed in partnership with Innate Pharma[1][3][5][6][7].
