Overview

SAR444336 is a site-specific, covalently-bound, monopegylated interleukin-2 (IL-2) engineered "Synthorin" developed using Expanded Genetic Alphabet platform technology. This approach expands the genetic code to create optimized biologics with improved properties. SAR444336 is designed as a non-beta IL-2 variant that selectively stimulates regulatory T cells (Tregs) by increasing reliance on the IL-2 receptor alpha chain subunit (CD25), thereby promoting immune tolerance and reducing inflammatory and autoimmune responses. Preclinical studies have shown that it can expand antigen-specific Tregs while limiting proliferation of NK and CD8+ T cells, making it an attractive candidate for controlling autoimmunity and inflammation. The drug is administered subcutaneously and is currently in Phase I clinical development for inflammatory diseases.
