SAR444656 (KT-474) is an investigational small molecule and heterobifunctional degrader that targets interleukin-1 receptor-associated kinase 4 (IRAK4) for degradation via the proteasome pathway. IRAK4 is a key signaling kinase downstream of toll-like receptors (TLRs) and interleukin-1 receptors, with a critical role in mediating inflammatory responses. By promoting degradation of IRAK4, SAR444656 blocks both its kinase and scaffolding functions, resulting in robust inhibition of pro-inflammatory cytokine signaling. The drug is in Phase 2 clinical trials for moderate to severe atopic dermatitis and hidradenitis suppurativa. SAR444656 is being developed by Kymera Therapeutics in partnership with Sanofi for inflammatory diseases[5][6][7][8].
