SAR445514 is a trifunctional anti-BCMA natural killer (NK) cell engager developed for the treatment of relapsed/refractory multiple myeloma and light chain amyloidosis. It is designed to simultaneously bind B-cell maturation antigen (BCMA) on tumor cells and activate NK cells by engaging both the activating receptors Natural cytotoxicity triggering receptor 1 (NKp46) and Low affinity immunoglobulin gamma Fc region receptor III-A (CD16a). This dual engagement facilitates the formation of an immunological synapse between NK cells and BCMA-positive tumor cells, leading to targeted tumor cell killing through enhanced NK cell activation and degranulation. The drug utilizes Sanofi’s proprietary CROSSODILE multi-functional platform in a Cross-Over-Dual-Variable-Domain (CODV) format for optimized activity[1][5][7]. It is administered subcutaneously in clinical trials as monotherapy.
