SAR445761 is an orally administered small molecule inhibitor of receptor-interacting serine/threonine-protein kinase 1 (RIPK1), a key signaling protein involved in mediating inflammation and necroptotic cell death. Developed by Sanofi in collaboration with the Medical Center - University of Freiburg, SAR445761 is currently being evaluated in Phase 2 clinical trials for the treatment of steroid-refractory overlap syndrome in patients with chronic graft-versus-host disease (cGvHD). By targeting RIPK1, the drug aims to inhibit the production of pro-inflammatory cytokines and prevent the programmed cell death that contributes to the systemic inflammation and tissue damage seen in GvHD. This targeted approach represents a potential therapeutic strategy for patients who do not respond adequately to standard corticosteroid therapy.
