SAR445877 is a bi-functional fusion protein drug candidate under development by Sanofi for the treatment of advanced solid tumors, including colorectal cancer, non-small cell lung cancer, metastatic hepatocellular carcinoma (HCC), gastric cancer, and gastroesophageal junction adenocarcinoma. The molecule consists of an anti-PD1 antibody fused to the cytokine IL-15. Its mechanism of action involves blocking PD-1 to enhance T cell activity while simultaneously stimulating the proliferation and activation of natural killer (NK) cells, cytotoxic T lymphocytes (CTLs), and memory T cells within the tumor microenvironment. This dual action aims to boost antitumor immune responses locally at tumor sites. SAR445877 is administered intravenously as a solution and is currently being evaluated in Phase 1/2 clinical trials both as monotherapy and in combination with other anticancer agents such as cetuximab[1][3][4][5][7].
