Overview

SAR446329 (also known as AMX-500) is a dual-masked bispecific T-cell engager antibody targeting prostate-specific membrane antigen (PSMA) and CD3. It is designed to direct cytotoxic T cells to PSMA-expressing tumor cells in metastatic castration-resistant prostate cancer (mCRPC). The drug uses a protease-cleavable masking technology to reduce off-tumor toxicity by activating the antibody preferentially within the tumor microenvironment. Originally developed by Amunix Pharmaceuticals and later advanced by Sanofi, it has been licensed to Vir Biotechnology for further development. The primary indication under investigation is metastatic castration-resistant prostate cancer[1][3][4][5][6][8].
