SAR448851 is an investigational TREM2 (Triggering Receptor Expressed on Myeloid cells 2) agonist being developed by Sanofi, in collaboration with Amgen, for the treatment of Alzheimer's disease. TREM2 is a transmembrane receptor primarily expressed on microglia, the resident immune cells of the central nervous system. It plays a critical role in microglial survival, phagocytosis, and the regulation of neuroinflammation. In Alzheimer's disease, microglial dysfunction is associated with the accumulation of amyloid-beta plaques and accelerated neurodegeneration. SAR448851 is designed to activate TREM2 signaling, thereby enhancing microglial-mediated clearance of pathological protein aggregates and potentially slowing disease progression. The drug is currently in Phase 1 clinical development, where it is being evaluated for safety, tolerability, and pharmacokinetics in patients with early Alzheimer's disease.
