Drug intelligence / Profile preview
Sardomozide (SAM486) is a potent, selective, and competitive small molecule inhibitor of S-adenosylmethionine decarboxylase 1 (AMD1/SAMDC), a key rate-limiting enzyme in the polyamine biosynthesis pathway. Originally developed by Ciba-Geigy (which later became Novartis) under the code name CGP 48664, sardomozide prevents the conversion of S-adenosylmethionine to decarboxylated S-adenosylmethionine, thereby depleting intracellular pools of spermidine and spermine. This depletion leads to cell cycle arrest and apoptosis in rapidly proliferating cells. Sardomozide has been evaluated in preclinical and early-phase clinical trials for various malignancies, including melanoma, non-Hodgkin lymphoma, and solid tumors, and continues to be investigated in preclinical combination regimens for neuroblastoma and non-small cell lung cancer (NSCLC).
