The saRNA COVID-19 vaccine is a self-amplifying RNA (saRNA) vaccine program developed by VLP Therapeutics (and its subsidiary VLP Therapeutics Japan) for the prevention of COVID-19 caused by SARS-CoV-2. The vaccine platform utilizes a self-amplifying replicon RNA delivered via lipid nanoparticles (LNPs). The initial candidate, VLPCOV-01, encodes a membrane-anchored receptor-binding domain (RBD) derived from the ancestral SARS-CoV-2 spike protein. To reduce reactogenicity, the platform was updated in VLPCOV-02 by incorporating a modified base, 5-methylcytosine (5mC), and updating the RBD to target the Gamma variant. Subsequent iterations include VLPCOV-04 (targeting Omicron XBB.1.5) and VLPCOV-05 (targeting Omicron JN.1), which have progressed to Phase 3 clinical trials in Japan as booster vaccines. The self-amplifying mechanism allows for robust and prolonged antigen expression at significantly lower doses compared to conventional mRNA vaccines.
