saRNA COVID-19 Variant Vaccine is an investigational self-amplifying RNA (saRNA) vaccine developed by VLP Therapeutics for the prevention of COVID-19 caused by SARS-CoV-2 variants. The vaccine utilizes a proprietary replicon platform derived from alphavirus vectors (such as Venezuelan equine encephalitis virus). Unlike conventional mRNA vaccines, saRNA encodes both the target antigen (SARS-CoV-2 spike protein) and a viral replicase complex. Once inside host cells, the replicase amplifies the RNA transcripts, enabling sustained and high-level antigen expression at significantly lower doses than standard mRNA vaccines. This dose-sparing effect is designed to reduce reactogenicity and manufacturing costs while inducing robust humoral and cellular immune responses. The program includes variant-adapted candidates such as VLPCOV-02 (Gamma variant-adapted) and VLPCOV-05 (Omicron JN.1-adapted).
