The saRNA Ebola vaccine is a self-amplifying ribonucleic acid (saRNA) vaccine candidate developed by Imperial College London for the prevention of Ebola virus disease. Utilizing a self-amplifying RNA platform derived from an alphavirus backbone (typically Venezuelan Equine Encephalitis Virus), the vaccine encodes both the Ebola virus glycoprotein (GP) antigen and a viral replicase complex. Upon intramuscular administration, the replicase enables the intracellular amplification of the antigenic RNA transcript, leading to sustained and high-level expression of the Ebola GP. This mechanism is designed to elicit robust humoral and cellular immune responses at significantly lower doses compared to conventional mRNA vaccines. The vaccine is currently being evaluated in a Phase I clinical trial (NCT05370014) alongside similar saRNA candidates for Marburg virus and Lassa fever, with funding from the Coalition for Epidemic Preparedness Innovations (CEPI).
