The saRNA Lassa fever vaccine is a self-amplifying RNA (saRNA) prophylactic candidate developed by Imperial College London to prevent Lassa fever. Utilizing an alphavirus-derived replicon (typically from the Venezuelan Equine Encephalitis Virus), the vaccine encodes the Lassa virus glycoprotein complex (GPC) antigen alongside a viral replicase. This self-amplification mechanism allows for significantly lower RNA doses compared to conventional mRNA vaccines while maintaining high levels of antigen expression and robust immune activation. The vaccine is currently being evaluated in a Phase I clinical trial (NCT05985798) in healthy adults, where it is administered intramuscularly in a two-dose schedule. This program is part of a broader effort, supported by the Coalition for Epidemic Preparedness Innovations (CEPI), to develop rapid-response vaccine platforms against high-priority viral threats, including Ebola and Marburg viruses.
