Saros First Agent is a preclinical-stage STING (Stimulator of Interferon Genes) agonist developed by Saro Therapeutics for the treatment of solid tumors, including melanoma. The therapeutic consists of a nanoparticle formulation that encapsulates cyclic dinucleotides (CDNs) along with manganese (Mn2+) ions. Manganese acts as a potent sensitizer and co-activator of the STING pathway, significantly enhancing the activity of CDNs and potentially allowing for lower dosing with improved efficacy. The nanoparticle is engineered to target the tumor microenvironment and activate innate immune cells, such as dendritic cells and macrophages. This activation triggers the production of Type I interferons and other pro-inflammatory cytokines, ultimately fostering a robust, systemic T-cell-mediated anti-tumor immune response.
