A combination vaccine comprising a recombinant protein subunit containing the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein, formulated with alum (aluminum hydroxide) and CpG 1018 as adjuvants. The RBD is a critical segment of the spike protein responsible for binding to the ACE2 receptor on host cells, making it a prime target for neutralizing antibody responses. CpG 1018 is a toll-like receptor 9 (TLR9) agonist that enhances vaccine-induced immunity. This combination is designed to elicit strong humoral and cellular immune responses against SARS-CoV-2, including multiple variants, by focusing immune responses specifically on the highly immunogenic RBD region. Such vaccines are being used or evaluated in multiple settings for the prevention of COVID-19, aiming to improve immunogenicity, breadth, and durability of protection compared to unadjuvanted protein subunits[1][2][4][6].
