Overview

Sasanlimab + tremelimumab is an investigational combination of two monoclonal antibodies developed for cancer immunotherapy. Sasanlimab is a humanized IgG4 monoclonal antibody that selectively targets and blocks the programmed cell death protein 1 (PD-1) receptor on T cells, preventing interaction with PD-L1 and PD-L2, and thus removing inhibitory signals that suppress T cell activation. Tremelimumab is a fully human IgG2 monoclonal antibody that blocks cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), another immune checkpoint receptor, enabling further activation of T cells against tumors. This dual checkpoint blockade is designed to enhance anti-tumor immune responses more effectively than monotherapy with either agent. Both agents are under investigation for use in various cancers, with sasanlimab most studied in bladder cancer, and tremelimumab approved (in combination with durvalumab) for unresectable hepatocellular carcinoma[1][2][4][6][8].
