SASPject PT4 is an investigational bacteriophage-based antibacterial agent from Phico Therapeutics that uses engineered, replication-defective phages to deliver a gene encoding a small acid-soluble spore protein (SASP) into target Gram-negative bacteria, specifically Klebsiella pneumoniae and Escherichia coli, including multidrug-resistant strains. Once expressed inside the bacterium, SASP binds broadly to bacterial DNA and shuts down essential cellular functions, leading to rapid bacterial death while minimizing the potential for resistance due to its non–sequence-specific DNA interaction. The product is designed for intravenous administration for treatment and prevention of systemic infections caused by these pathogens and exemplifies Phico’s SASPject platform as a “first-in-class” non-traditional antibacterial approach aimed at carbapenem-resistant Enterobacteriaceae and other difficult-to-treat infections.[4][5][8][10][11][12]
