Overview

SAT-3153 is a first-in-class, orally administered small molecule therapeutic developed by Satellos Bioscience. It is designed to restore the innate muscle regeneration process in patients with Duchenne muscular dystrophy (DMD) by inhibiting adaptor-associated kinase 1 (AAK1), a protein kinase involved in the Notch signaling pathway that regulates muscle stem cell polarity and division. Unlike current DMD therapies, SAT-3153 acts independently of dystrophin and exon mutation status, aiming to enhance muscle regeneration and function by modulating stem cell polarity. Preclinical studies have shown that SAT-3153 increases muscle force and promotes new progenitor muscle cell formation in animal models of DMD. The drug has received Orphan Drug Designation and Rare Pediatric Disease Designation from the U.S. FDA for DMD but remains at the preclinical stage as a backup candidate to Satellos’ lead compound, SAT-3247[1][2][3][6][7].
