Satraplatin is an orally administered, third-generation platinum-based chemotherapeutic agent developed as the first oral platinum compound for cancer treatment. Unlike other platinum analogues such as cisplatin and oxaliplatin, which require intravenous administration, satraplatin can be taken orally. Its mechanism of action involves forming highly reactive, charged platinum complexes that bind to nucleophilic groups in DNA, inducing both intrastrand and interstrand DNA cross-links as well as DNA-protein cross-links. These interactions inhibit cell division and induce apoptosis in cancer cells. Satraplatin has shown activity against a variety of malignancies including prostate cancer (especially castrate-resistant metastatic prostate cancer), lung cancer (including non-small cell lung cancer), ovarian cancer, breast cancer, head and neck cancers, and some hematological malignancies. It was primarily investigated for use in patients with advanced or hormone-refractory prostate cancer who had failed previous chemotherapy regimens[1][2][4][5][7][10].
